Cleared tissue

Cleared and expanded tissues

We recently demonstrated SCAPE 2.0's ability to image large, cleared and expanded tissues. Instead of using SCAPE's scanning mirror, here we translated the samples at constant speed, grabbing volumes up to 10 mm x 1 mm x 400 um in each sweep. We can then stitch these volume blocks back together to form a complete, high resolution volume at speeds 100's of times faster than point-scanning microscopes.

See our new SCAPE 2.0 paper for more details:

SCAPE 2.0 image of an mCubic cleared mouse brain (Thy1-GFP). The 8.4 x 9 x 0.4 mm xyz volume was acquired within 4 minutes with 1 x 1.37 x 1.14 micron per pixel sampling density. Rendered using Imaris (Bitplane). Sample provided by Pavel Osten Lab.
SCAPE 2.0 image of a 4x expanded mouse spinal cord, was acquired within 100 seconds. Rendered using Imaris (Bitplane). Sample provided by Dawen Cai Lab.
